Dye Blobs



These off scale peaks consist of unincorporated dye terminators interacting non-covalently with an unknown. This particular blob pattern and location at 50 to 130 bases persists in SDS untreated sequencing reactions. Diminished blobs may still occur after SDS and be problematic in low signal sequences. Our SDS treatment is applied to each sample after PCR as follows: 0.2% SDS, final concentration, and heat the samples to 95°C for 5min.
