Chromatogram Tips
- Chromatogram quality should always be considered before using a sequence.
- Multiple sequencing problems may occur simultaneously.
- Black G peaks are artificially depressed when flanking green A peaks.
- The first 20 bases off of the 3'end of the primer do not make it past purification.
- The first 30 bases of a chromatogram can not be trusted. Mass differences between the bases weigh more heavily at these short lengths.
